RESUMO

Covalent organic framework (COF) nanospheres with controlled size of ∼500 and ∼1100 nm were successfully prepared by adjusting the HOAc amount in the synthetic system. The as-synthesized COFs have large conjugate aromatic skeleton, excellent stability, abundant pore, and uniform morphology. These advantages of COFs are benefit for immobilizing aptamers to fabricate the targeted electrochemical aptasensor. The commonly used oxytetracycline (OTC) is an analytic model to explore the sensing performance of the COF-based aptasensor, indicating that the smaller COF (∼500 nm) is more conducive to acquiring the sensitive sensor than that of the larger COF (∼1100 nm). Moreover, the limitation of detection of the COF (∼500 nm)-based aptasensor is calculated to be 7.4 fg mL-1 using the response impedance signal. Additionally, the aptamer-based biosensor has fine reproducibility, good stability, excellent specificity, and available usability even in real samples.

Background: Gestational diabetes mellitus (GDM) is a common metabolic disorder in pregnancy. The incidence rate is increasing year by year, which seriously threatens the safety of maternal and infant. Obesity is a vital factor in inducing GDM. Pregnant women with GDM account for a large proportion of overweight and obese pregnant women. Our study aimed to explore the potential mechanism of differential metabolites on inflammation and find the intervention and management methods for GDM in overweight and obese pregnant women. Methods: Umbilical cord blood samples and placenta were collected from normal weight pregnant women with GDM (control group) and overweight and obese pregnant women with GDM (obesity group) for a comparative study. Serum inflammatory factors IL-10, TNF-α, IL-6, lipopolysaccharide (LPS), and TLR4 expression were detected by ELISA. The expression levels of BCL-2 and caspase-3 were measured by Western blot. TUNEL staining was used to observe the apoptosis of placental villi. KEGG combined with metabolomics was used to compare the differences of metabolic maps between the two groups. Results: Compared with the control group, the level of anti-inflammatory factor IL-10 in the cord blood was decreased in the obesity group, while the levels of proinflammatory factors TNF-α, IL-6, and LPS were increased. In the placental tissues, the obesity group had higher concentrations of LPS, TLR4, and caspase-3 and lower concentration of BCL-2. Placental villi in the obesity group were more likely to undergo apoptosis than the control group. Correlation analysis showed that the above metabolite concentrations were negatively correlated with TNF-α or LPS. Conclusion: Metabolites could control obesity in the process of controlling the occurrence and development of inflammation.

Benzo(a)pyrene (BaP) is a polycyclic aromatic hydrocarbon that specifically causes cancer and is widely distributed in the environment. Poly (ADP-ribosylation), as a key post-translational modification in BaP-induced carcinogenesis, is mainly catalyzed by poly (ADP-ribose) glycohydrolase (PARG) in eukaryotic organisms. Previously, it is found that PARG silencing can counteract BaP-induced carcinogenesis in vitro, but the mechanism remained unclear. In this study, we further examined this process in vivo by using heterozygous PARG knockout mice (PARG+/-). Wild-type and PARG+/- mice were individually treated with 0 or 10 µg/m3 BaP for 90 or 180 days by dynamic inhalation exposure. Pathological analysis of lung tissues showed that, with extended exposure time, carcinogenesis and injury in the lungs of WT mice was progressively worse; however, the injury was minimal and carcinogenesis was not detected in the lungs of PARG+/- mice. These results indicate that PARG gene silencing protects mice against lung cancer induced by BaP inhalation exposure. Furthermore, as the exposure time was extended, the protein phosphorylation level was down-regulated in WT mice, but up-regulated in PARG+/- mice. The relative expression of Wnt2b and Wnt5b mRNA in WT mice were significantly higher than those in the control group, but there was no significant difference in PARG+/- mice. Meanwhile, the relative expression of Wnt2b and Wnt5b proteins, as assessed by immunohistochemistry and Western blot analysis, was significantly up-regulated by BaP in WT mice; while in PARG+/- mice it was not statistically affected. Our work provides initial evidence that PARG silencing suppresses BaP induced lung cancer and stabilizes the expression of Wnt ligands, PARG gene and Wnt ligands may provide new options for the diagnosis and treatment of lung cancer.

Poly (ADP-ribosylation) is a key post-translational modification (PTM), and poly (ADP-ribose) glycohydrolase (PARG) is the main enzyme that hydrolyzes poly (ADP-ribose) in eukaryotic organisms. Our previous findings suggested that knockdown of PARG attenuates benzo(a)pyrene (BaP) carcinogenesis. However, the mechanisms underlying PARG-mediated protective effects remain limited. In this study, the expression levels of histones were analyzed by Western blotting and immunofluorescence. Histone H2A levels were abnormally decreased by BaP-induced carcinogenesis, but were maintained by knockdown of PARG in the 16HBE human bronchial epithelial cell line. The interaction between poly (ADP-ribose) and H2A was confirmed by co-immunoprecipitation. PARG-related modifications in H2A were profiled by immune antibody enrichment coupled with mass spectrometry. H2AK5ac, H2AK9ac, H2AK13ac, H2A.ZK4K7K11ac, and H2AK9me were expressed in BaP-transformed 16HBE (BTC-16HBE) cells, but were not detectable in normal 16HBE or BaP-transformed 16HBE cells with knockdown of PARG (BTC-shPARG). Further verification by Western blotting indicated that H2AK9me was elevated in BTC-16HBE cells but decreased in BTC-shPARG cells. These findings suggest that knockdown of PARG protects against BaP-induced carcinogenesis in 16HBE cells by downregulating H2AK9me. Our in vivo studies confirmed that PARG silencing decreased H2AK9me levels, thereby countering the carcinogenic teratogenic effects induced by BaP.

OBJECTIVE: To develop a self-administered risk questionnaire for common nutrition-related diseases in middle school students. METHODS: Two phases were conducted to develop the questionnaire: scale development and validation. Phase 1 included 7 steps: (1) determining the objective, theoretical framework, principles and format for indicator generation; (2) setting up the preliminary indicator pool; (3) selecting indicators and forming pilot questionnaire through focus groups; (4) testing the pilot questionnaire; (5) further correcting the questionnaire using expert consultation; (6) choosing indicators again using good-poor analysis; and (7) shaping the final questionnaire. Phase 2 consisted of: (1) using the Pearson correlation coefficient to assess test-retest reliability; (2) using the Cronbach's alpha coefficient to assess the internal consistency reliability; (3) using the feedback from field investigation to assess face validity; and (4) using explanatory factor analysis to assess construct validity. Students from 96 classes were selected at random in Hunan Province as the field test samples using stratified sampling and cluster sampling. And the students from 4 out of the 96 classes were chosen again to serve as the test-retest samples. We used Epidata 3.0 to build the database and SPSS 11.0 to analyze the data. RESULTS: A brief self-administered risk questionnaire for common nutrition-related diseases in middle school students with 12 items being formed after Phase 1. Good-poor analysis showed results from t tests for each item were statistically significant (P<0.05). The Pearson correlation coefficient was 0.76 (P<0.05) and the Cronbach's alpha coefficient was 0.56. The questionnaire was accepted by the students participating in the field test. Four common factors were extracted using explanatory factor analysis, accounting for 50.18% of the total variation. CONCLUSION: The brief self-administered risk questionnaire for common nutrition-related diseases in middle school students is reliable and valid.

OBJECTIVE: To study the prevalence of internet addiction disorder (IAD) in middle school students of Hunan and to explore its risk factors. METHODS: 5760 middle school students and their parents were sampled at random in Hunan province using two-stage sampling (stratified sampling and cluster sampling) method. The ten-item diagnosis tool for IAD, self-rating depression scale, self-rating anxiety scale, interpersonal sensitivity subscale of symptom checklist, family assessment device, parenting locus of control scale, and self-developed questionnaire were employed together to gather related data. 95% confidence interval (CI) was used to describe the prevalence of IAD. Chi-square and logistic regression tests were employed respectively to compare the differences of IAD prevalence among different subpopulations and to explore the possible influential factors. RESULTS: The overall prevalence of IAD in middle school students of Hunan was 5.52%, with 95% CI as 4.84-6.20. IAD prevalence rates between males and females, being or not being monitored in the class and among different grades, showed significant differences (P < 0.05). No statistical difference was found between single child or having siblings in the family, being at key middle school or ordinary middle school, or within residential areas, (P > 0.05). Gender, tired of going to school, anxiety, interpersonal sensitivity, peer influence in haunting at internet bars, father's education level, the number of recreational settings in the community etc. were the influencing factors of IAD, with ORs as 0.281, 3.469, 2.318, 1.710, 1.877, 1.456, 1.273 and 0.726 respectively (P < 0.10). CONCLUSION: The prevalence of IAD in middle school students of Hunan was moderate compared to the reported prevalence rates in other provinces. Gender and peer influence in haunting at internet bars and other 6 factors were suggested to be correlated with IAD events.
